The co-culture of endothelial cells, pericytes and astrocytes mimick the anatomical situation in vivo. In the presence of both pericytes and astrocytes rat brain endothelial cells expressed enhanced levels of tight junction (TJ) proteins occludin, claudin-5 and ZO-1 with a typical localization at the cell borders. Pericytes were then seeded (1.5 × 10 4 cells/cm 2) on the bottom side of polycarbonate inserts and placed upside down.

Brain microvascular cells such as brain microvascular endothelial cells (BMEC), pericytes, and astrocytes are major components of the blood–brain barrier (BBB). The BBB regulates substance transport from the blood into the brain and is involved in the progression of many neurological diseases. Most studies have focused on neurons while ignoring the important roles of other cells of the neurovascular unit, such as astrocytes and pericytes. Astrocytes are important for the development and maintenance of the blood–brain barrier, brain homeostasis, structural support, control of cerebral blood flow and secretion of neuroprotective factors.
